Abstract It is clearly established that the immune system can affect cancer response to therapy. However, influence of tumor microenvironment (TME) on cells not completely understood. In this respect, alternative splicing increasingly described system. Here, we showed TME, via a TGFβ-dependent mechanism, increased events and induced expression an isoform IRF1 transcription factor (IRF1Δ7) in Th1 cells. We found SFPQ (splicing factor, proline- glutamine-rich) was responsible for IRF1Δ7 production. also showed, both mice humans, altered full-length transcriptional activity Il12rb1 promoter, resulting decreased IFNγ secretion Thus, inhibiting could potentiate antitumor responses.
